Set the speed to a negative value (-100% for normal countdown). Add days, hours, minutes, and/or seconds to the start time.
The colons are customizable with Count Pro or Type. Change the actual text of the text layer (i.e. “.”). Then toggle on “Custom Colons.” The text you typed will replace the colons.
Make sure your expression engine is set to JavaScript.
